Metabolic oscillations in yeast

The baker's yeast Saccharomyces cerevisiae, when grown in continuous cultures under the right con- ditions, shows spontaneous metabolic oscillations that affect not only gas exchange rates and metabolite concentrations, but also the expression of a large number of genes. Here I integrate time-dependent metabolite concentrations, oxygen uptake rates, carbon dioxide and ethanol excretion rates, as well as thermodynamic data into a large network model of yeast metabolism. By fitting a linearised kinetic model to the data, and by assuming that metabolic dynamics is entirely driven by the time-dependent oxygen uptake rate, known from experiments, I determine metabolic fluxes, concentrations of non-measurable metabolites, and local metabolite concentrations in cell compartments (cytosol and mitochondria) along the metabolic cycle.
